Dissertation note: Thesis (PH.D.) - Cairo University - Faculty Of Agriculture - Department Of Plant Pathology and Physiology Summary: Total DNA for ten begomoviruses were successfully extracted from tissues of healthy and infected plant grown in field and greenhouse by applying CTAB methodThe PCR was used in the detection of begomoviruses under study in tissues of infected plants growing in the fieldThe degenerate primers (AV - core and AC - core) for begomoviruses were able to amplify products of 576 - 579 bp of core coat protein of all viruses under study except for CLCMVAlso results indicated the efficiency of IC - PCR in the detection of OLCV and TYLCV in sap extracted from infected plants growing in the field using 1 / 500 TYLCV - CP antiserumNon - radioactive hybridization technique was employed in order to detect and differentiate between begomoviruses under study in both of infected and uninfected plant tissuesThe non radioactive - labeled probe of SLCV was prepared using PCR amplification with degenerate primers of core coat protein of geminivirusesSLCV labeled probe was used in squash , dot , and Southern blot hybridization techniquesResults indicated a close sequence homology between SLCV and TYLCV , and a moderate sequence homology with each of OLCV , HLCrV and OYVVIn addition , results predicted that DNA sequences of the other viruses and SLCV are less conserved
