Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Science- Department of Biochemistry Summary: This work has been carried out to assess the cytotoxic potential of glufosfamide (GLU) in a hepatocellular carcinoma cell line paradigm; namely (HepG2), in comparison to its parent aglycone ifosfamide (IFO). HepG2 cells were used for assessment of cell viability and apoptotic cell death using MTT technique and flowcytometric analysis, respectively. Cell lysate was prepared to determine Bcl2 and caspase-3 gene expression (by Real time PCR), caspase-9 enzyme content (by ELISA) and ATP concentration (by luciferease-luminescence assay technique). The current study was extrapolated to assess any possible hepatotoxicity for glufosfamide and its parent compound; ifosfamide. The biochemical test battery entailed assessment of liver transaminases activities, oxidant/anti-oxidant status as manifested by measurement of tissue GSH and MDA contents, and both catalase and SOD activities. Histopathological examination of liver sections was achieved to confirm hepatotoxicity
