Liver cancer is the fifth most common cancer worldwide and the third most deadly. This study aimed to explore the association of LAPTM4B gene polymorphism with HCC in Egyptian patients and whether this genetic polymorphism is associated with the clinicopathological characteristics of the disease. The study included 70 patients with hepatocellular carcinoma. And 50 healthy subjects serving as control. Physical and clinical examinations were done to all subjects. 10 ml of blood were collected from each subject. The following was done; Liver function tests, CBC, DNA extraction from whole blood, genotyping of LAPTM4B gene by PCR, agarose gel electrophoresis and quantitation of protein level of LAPTM4B by ELISA. LAPTM4B *2 / 2 was associated with a significant increased risk of HCC compared with LAPTM4B*1 / 1 (p = 0.006). It is also found that the frequency of allele *2 was higher in HCC (47.9%) than in controls [p < .05]. There was higher level of LAPTM4B protein among HCC patients genotype 2* / 2* (1680.82 ± 654.89) than those have genotype 1* / 2* (1216.25 ± 650.89) and 1*/1* (1144.80 ± 659.98).
