Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Medicine - Department of Clinical and Chemical Pathology Summary: Background: Hepatocellular carcinoma is one of the most common and lethal malignant tumors worldwide. MicroRNAs are involved in the epigenetic mechanism{u2019}s implicated in HCC molecular pathogenesis. Measuring the levels of the circulating miRNAs to be used as biomarkers for malignancies has recently been investigated in the hope of identifying unique miRNA signatures in human tumors. Two clusters of microRNAs have been selected in this study; Let7-a-1, let-7d-1, let-7f-1 cluster and miR143/145 cluster. Aim of the study: To identify miRNA let-7 a-1, d-1, f-1 cluster and miRNA143/ 145 cluster gene expressions in HCC patients. Methods: The study included 40 Hepatocellular carcinoma patients, 40 chronic HCV patients divided into 2 subgroups, 20 patients with liver cirrhosis and 20 patients without liver cirrhosis, and 40 apparently healthy subjects as a control group. They were subjected to routine laboratory investigations and gene expression analysis for miRNA let-7 a-1, d-1, f-1 cluster and miRNA 143/ 145 cluster using quantitative SYBR Green reverse-transcription Real-Time polymerase chain reaction
