Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Pharmaceutical Chemistry
The study in this thesis included new analytical methods for the determination of some drugs and prodrugs in combination with their degradation products or active metabolites.Drugs cited in the thesis aresome antiviral drugs, antidiabeticdrugs and drugs used for treatment of inflammatory bowel disease. These methods include RP-LC method for simultaneous determination of seven antiviral drugs, the quantification was achieved at 254 nm for (acyclovir, ganciclovir, penciclovir and valaciclovir hydrochloride) and at 220 nm for (lamivudine, oseltamivir phosphate and ribavirin). Moreover, bioanalytical method for simultaneous determination of lamivudine and penciclovir in spiked human plasma wasdeveloped using valaciclovirHCl as internal standard. Validation of bioanalyticalmethod was performed according to European Medicines Agency guidelines.RP-LC method for determination of daclatasvirdihydrochloridewas developed and the study of its degradation behavior was investigated.Peak purity was checked for principle drug and its alkaline induced degradation product and the pathway of alkaline hydrolysis of daclatasvirdihydrochloride was suggested by LC/MS
