Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Biochemistry Summary: miRNA-34a was reported as urinary marker for early detection of diabetic nephropathy. In this work, a simple colorimetric gold nanoparticle (AuNP) assay was developed for rapid and sensitive detection of unamplified urinary exosomal miRNA-34a based on the surface plasmon resonance (SPR) property of AuNPs. The nano-assay uses citrate-capped AuNPs, specific probe targets miRNA-34a and NaCl as an aggregation inducer. miRNA-34a-probe hybrid stabilizes AuNPs against salt induced aggregation. The method performed in totally extracted urinary exosomal microRNA from normal and type 2 diabetic subjects. The AuNP assay results were compared with conventional qRT-PCR results.The sensitivity and specificity for qualitative detection of urinary miRNA-34a using the AuNP assay were 81.08% and 69.6% respectively.The sensitivity of detection of urinary miRNA-34a by qRT-PCR was (83.8%), while specificity was (93.3%).The method is simple, cost-efficient, has a short turnaround time and requires minimal equipment and personnel for urinary miRNA-34a detection
