Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Pharmacology and Toxicology Summary: One of the most lethal cancers of the 20th century is lung cancer which is causing up to 3 million deaths annually worldwide.Therefore, management of lung cancer needs searching for new chemopreventive agents. This work was designed to inspect the chemopreventive potential of Synadenium grantii extracts on lung carcinogenicity induced by Ben-zo(a)pyrene [B(a)P] in male albino western rats. This target was undertaken through preparing different extracts (water, methanol, ethanol and chloro-form) from branches and leaves of S. grantii for screening their effect on lung cancer cell line (A549), then the most active extract was used for com-bating lung cancer in animal model. The in vitro results revealed that, the methanolic extract was found to be the most active extract against A549 cells. The methanolic extract of S. grantii has a notable cytotoxicity activity (IC50: 4.30±0.44 og/ml) against A549 cells more than the other extracts and its activity was close to the cytotoxicity of standard drug, doxorubicin (IC50: 3.50±040 og/ml) due to the presence of high flavonoids content. After the determination of S. grantii safe dose (305mg/Kg b.w.), which represents 1/10 of lethal dose, in the in vivo experiment, rats were divided into five groups (10 animal each)
