Diphenhydramine hydrochloride is one of the famous first generation H1 antihistaminic drugs that are used in the treatment of skin allergy. It reduces the actions of histamine in the body by competitive blockade of histamine H1 receptor sites on tissues. When orally taken, diphenhydramine hydrochloride is easily able to penetrate the blood brain barrier and occupy H1 receptor sites in the brain, resulting in fatigue, drowsiness, and performance impairment. Meanwhile conventional topical gels of diphenhydramine hydrochloride showed less patient compliance due to increased frequency of application on the skin to reach better therapeutic effect. Diphenhydramine hydrochloride loaded nanoparticles were prepared using different natural polymers and simple techniques for preparation. These nanoparticles could provide targeted therapy with enhanced skin delivery and prolonged anti-allergic effect .Consequently; this will lead to improvement in patient complianceThe aim of this work: 1. Preparation of diphenhydramine hydrochloride loaded nanoparticles containing adequate amount of drug to achieve better therapeutic effects than conventional topical gel preparations. 2. Prolongation of the therapeutic effect of diphenhydramine hydrochloride loaded nanoparticles which lead to decreased frequency of application of the drug in comparison to conventional topical gel preparations. 3. Improve patient compliance. Thus the work in this thesis was divided into three chapters: Chapter 1: Preparation and evaluation of diphenhydramine hydrochloride loaded chitosan nanoparticles. Chapter2: Preparation and evaluation of diphenhydramine hydrochloride loaded alginate/chitosan nanoparticles. Chapter3: In-vivo performance of the selected diphenhydramine hydrochloride loaded nanoparticles
