Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Science - Department of Botany and Microbiology Summary: Human Granulocyte colony-stimulating factor (hG-CSF) is a growth factor or cytokine produced by a number of different tissues to stimulate the bone marrow to produce granulocytes and stem cells. In our study we aimed to optimize production (expression) of recombinant human G-CSF protein by two different strategies. The first one is based on the isolation of natural human G-CSF mRNA from polymorphonuclear cells (PMNCs). The second one is based on the synthesis of hG-CSF gene based on the GenBank published sequence. The hG-CSF open reading frame (ORF) of both strategies were amplified and cloned into pET-3a expression vector and transformed into BL21 DE3 E. coli cells for expression of recombinant hG-CSF protein and was successfully expressed and tested by Western blot. The results of this work indicate that successful expression of recombinant hG-CSF protein by two strategies which confirmed by the molecular weight and the immunogenicity against the hG-CSF antibodies
