This investigation was carried out during the period from 2010 until 2014 in Central Lab of Plant Physiology, Faculty Agriculture Cairo University Giza, Egypt. This work was conducted to find suitable protocol to increase the productivity of Hyoscyamus muticus active ingredients (hyoscyamine and scopolamine) through hairy roots technique using gene transformation with different strains of Agrobacterium rhizogenes. Optimization of callus culture conditions from leaf explants of H. muticus as well as comparing the alkaloids (hyoscyamine and scopolamine) concentrations from different sources, i.e. three clones of hairy root cultures, callus cultures and intact normal H. muticus plants
