In the present study it was found that amiodarone administration in rats by gastric gavage tube resulted in changes in thyroid gland. These changes were in hormonal levels, histological and immunohistochemical changes. These changes were improved by treating rats with stem cells. Also, adding melatonin to the stem cells culture was found to have further beneficial effect. The present study was carried out to investigate the structural and functional changes in the thyroid gland following the administration of amiodarone, and to assess the possible role of stem cells and melatonin supplement in minimizing these changes in the gland of adult male albino rat as an animal model. On the other hand, malondialdehyde levels in thyroid tissue expressed a highly significant increase in groups III and IV compared to control group, while group V showed increased in only a significant manner than the control. Meanwhile, group IV and V showed statistically significant decrease from Group III. Also, group V decrease was insignificantly different from that of group IV. The PCR results showed highly significant increase of both IL-6 and VEGF in group III compared to the control one. Groups IV and V showed a highly significant decrease compared to group III
