Colorectal cancer is the third most common cancer, but it is the second most deadly cancer worldwide. In Egypt, Colorectal cancer is Egypt's seventh most prevalent cancer, accounting for 3.47 percent of male cancers and 3% of female cancers. Jasonia Montana is one of the most prevalent therapeutic herbs. The bio medicinal action of Jasonia montana is due to a variety of active compounds that combine to form the complex impact of the plant. Quercetin is one of the important flavonoids that exert antiproliferative effects on cancer cells mediated by various mechanisms of action including alterations in gene expression of oncogenes, tumor suppressor genes. MiRNAs are short, non-protein-coding RNAs 22 nucleotides in length that are known to alter gene expression at a post-transciptional level. Here, we investigated the potential efficacy of jasonia Montana to trigger colon cancer cells toward differentiation. In this study we fulfilled miRNAs& their targeted genes related to differentiation pathways such as NOTCH, PI3K/AKT & pluroipotancy gene SOX2 in both CACOO2&HCT116 colon cell lines and then evaluate stem cell marker CD44& differentiation marker CK20 to ensure the efficacy of jasonia Montana toward differentiation. Using syber green RT-qPCR , both miR-21& miR-10B were shown to be significantly downregulated While their targeted gene PTEN (which included in PI3K/AKT pathway) shown to be significantly upregulated . On the other hand, miR-34a was significantly up regulated &its targeted gene NOTCH1 was down regulated,miR-145 was upregulated while its targeted gene SOX2 was down regulated. Finally, CD44 as stem cell marker was downregulated while CK20 as differentiation marker was upregulated. In conclusion, jasonia Montana may be used as
