| Summary, etc. | In this thesis we prepared some Mn(I) and Ru(II) photo-activatable CORMs (photoCORMs) with different mono and bidentate ligand systems as CO pro-drugs. The complexes were fully characterized using different analytical and spectroscopic tools including single crystal X-ray diffraction for five complexes. The dark stability, CO photo-induced releasing properties and solvatochromism features were examined. Time-dependent density functional theory computations were used to understand more about the origin of the observed lowest energy transitions. The cytotoxicity of the titled photoCORMs against different malignant and normal cell lines were examined under both dark and illumination conditions. The titled complexes are either water-soluble or soluble in DMSO/H2O mixtures. The prepared photoCORMs released CO upon exposure to 365−468 nm LED. Some of the prepared photoCORMs exhibited cytotoxicity against different malignant cell lines, while others showed no observed cytotoxicity. This indicates that the iCORM (CO-depleted CORM) may play a role in the cytotoxic effect of the complexes. The Mn complex featuring N,N-bidentate ligand showed dose-dependent cytotoxicity against MDA- MB-231 and HEK-293T cells and enhanced the cytotoxicity of the drug paclitaxel in the event of resistance. The mono-carbonyl Ru(II) complex showed the highest cytotoxicity against leukemia with IC50 value of 7.13 ± 0.8 μM in the dark. |
