Summary, etc. Avian pathogenic Escherichia coli (APEC) is a ubiquitous poultry disease affecting the global poultry industry's economics. APEC shares numerous similarities and commonalities in serotypes, phylogroups, virulence genes, disease patterns, in vivo transcriptome of murine urinary infection model and chicken challenge model with human uropathogenic E. coli (UPEC). The research aims to detect the shared virulence factors, antimicrobial resistance (AMR) patterns, and extended-spectrum β-lactamase between APEC and UPEC isolates from poultry and humans in Egypt.
Materials and Methods: Livers, lungs, hearts, and spleens of chicken carcasses were sampled from 35 poultry farms. In addition, a total of 17 human urine samples were collected. Isolation and identification were carried out using conventional methods and biochemical tests. The detection of shared virulence factors of UPEC and APEC by polymerase chain reaction PCR and sequencing. Antimicrobial susceptibility testing according to the Clinical Laboratory Standards Institute Guidelines was measured. Double-disk confirmation test was performed for the extended-spectrum β-lactamase test.
Results: twenty nine isolates were retrieved from 52 chicken organs and human urine samples (55.7%). The resistance rate for all isolates was Amoxicillin/clavulanic acid AMC (100%), AMP (93.1%), CAZ (89.7%), and CTX (82.8%). While CTR was (51.7%), NIT (34.5%), CN (24.1%), and COL (27.6%). Although APEC isolates showed AMC (100%), AMP (100%), CAZ (100%), CTX (94.4%), TE (94.4%), SXT(94.4%), C (100%), DO(88.9%), NA(88.9%), CIP(72.2%), CTR (83.3%), NIT (55.6%), GEN (33.3%), COL (38.9%). Only 15/29 (51.7%) of isolates showed ESBL-positive tests. In the present study, 28/29 (96.5%) E. coli isolates revealed multidrug resistance (MDR). The iss gene (20.69%) was the highest in frequency, other virulence genes papC, and vgrG1 were found lower (6.9% and 3.45% respectively).
Conclusion: APEC multidrug resistance high prevalence to all antibiotics for poultry contributes to therapeutic failure. In addition, it can act as a reservoir of human infection by multidrug-resistant pathogens. Furthermore, the results suggest the zoonotic potential of some APEC strains.
520 ## - SUMMARY, ETC.
Summary, etc.
AMC (100%)، AMP (93.1%)، (89.7%)CAZ ، (82.8%)CTX.
