| Summary, etc. | Pyridazinone derivatives have a growing interest in synthesis because <br/>of their biological importance. They exhibit a wide variety of biological <br/>activities, including antimicrobial, analgesic, antidepressant, anticancer, anti-<br/>inflammatory, vasodilator, and many other types of activities. Cardiovascular <br/>diseases are the first cause of death worldwide; therefore, there are many <br/>strategies to combat this disease. Among these is the use of vasorelaxants. <br/>So, twenty-eight new 2-phenyl-3,6-pyridazinedione derivatives have <br/>been prepared and screened for their vasorelaxant performance by inhibition <br/>of the PDE-5 enzyme. A molecular design was used to test the binding <br/>technique of the compounds with the phosphodiesterase-5 (PDE-5) receptor. <br/>Results obtained from docking considerations were extremely compatible <br/>with those obtained through biological screening. Fundamentally, derivatives <br/>4b, 4g, 4h, and 11a inhibition of PDE-5 were promising with potent IC50 <br/>values = 25, 53, 22, and 42 nM, respectively in comparison with Sildenafil <br/>(IC50 = 16 nM). Furthermore, the four most active compounds were safe for <br/>the normal fibroblast cell line WI-38. Also, 4f, 4h, 4j, 10d, and 11d <br/>demonstrated effectively same anti-proliferative effects on the aortic cell line <br/>as Sildenafil. |
