This study evaluated glutamic acid (Glu) production by probiotic bacteria
and the impact of various stimulants. Seventy samples, including, yogurt, cottage
cheese, and raw milk were analyzed. Bacillus subtilis and Enterococcus faecium
were the main probiotic bacteria identified. Seven strains of E. faecium and one
strain of B. subtilis were isolated and confirmed through PCR with specific primers
Liguid chromatography mass spectrometry mass spectrometry (LC-MS/MS) was
used for qualitative Glu estimation. The study quantified extracellular (ECG) and
intracellular glutamic acid (ICG) produced by untreated strains using LC-MS/MS
To stimulate Glu-responsible genes, sucrose-supplemented medium and UV light
exposure were employed. UV light was most effective for ECG and ICG production
in B. subtilis and for ICG production in E. faecium. Sucrose supplementation
significantly enhanced ECG production in E. faecium.
The study determined that exposing B. subtilis to UV light and utilizing
sucrose-supplemented medium for E. faecium are the best methods for achieving
cost-effective and safe production of Glu.
قامت هذه الدراسة بهدف تقييم إنتاج حمض الجلوتاميك بواسطة البكتيريا الحيوية وتأثير المنشطات المختلفة. تم تحليل سبعين عينة، بما في ذلك الحليب الخام والجبن القريش والزبادي. كانت باسيلس ساتلس وإنتيروكوكس فيشيم من البكتيريا الحيوية الرئيسية التي تم تحديدها. تم عزل سبع سلالات من إنتيروكوكس فيشيم وسلالة واحدة من باسيلس ساتلس وتأكيدها من خلال تفاعل البوليميراز المتسلسل باستخدام بادئات محددة. تم استخدام جهاز الإل سى ماس ماس لتقدير إنتاج حمض الجلوتاميك النوعي. قامت الدراسة بقياس كمية حمض الجلوتاميك خارج الخلية وداخل الخلية المنتجة بواسطة سلالات غير معالجة باستخدام .تم استخدام جهاز الإل سى ماس ماس. لتحفيز الجينات المسؤولة عن إنتاج حمض الجلوتاميك ، تم استخدام وسط مكمل بالسكروز والتعرض للأشعة فوق البنفسجية. كان ضوء الأشعة فوق البنفسجية أكثر فعالية لإنتاج كمية حمض الجلوتاميك خارج الخلية وداخل الخلية في باسيلس ساتلس ولإنتاج حمض الجلوتاميك داخل الخلية في وإنتيروكوكس فيشيم. عززت مكملات السكروز بشكل كبير إنتاج ولإنتاج حمض الجلوتاميك خارج الخلية في إنتيروكوكس فيشيم. وخلصت الدراسة إلى أن التعرض للأشعة فوق البنفسجية لـلباسيلس ساتلس ووسط مضاف إليه السكروز لـلإنتيروكوكس فيشيم هو الأمثل لإنتاج الجلوكوز بشكل آمن واقتصادي.
