Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Microbiology and Immunology Summary: The production of microbial penicillin G acylase (PGA) was investigated by six bacterial and two actinomycetal isolates. The PGA production by the chosen bacterial isolate, Arthrobacter viscosus NRRL B-1973, was optimized, appling statistically designed experiments. First, the Plackett-Burman design (PBD) was used to screen the effect of 19 factors, including medium components and culture conditions. Among the nineteen tested factors, only six were proven to be significant. These were; tryptone, arginine, maltose and dextrin, which exerted a significant positive effect on the PGA production, while both MgSO4 and NaCl offered significant negative effects
