Summary: Background: Testicular dysfunction is one of the complications of type 2 diabetes mellitus (T2DM) in men, this is illustrated by androgen deficiency, deterioration of sperm quality and the impaired fertility. Hence, trials to restore the testicular functions using pharmacological and non-pharmacological approaches are of a great interest. Aim of work: The present study was designed to compare the possible therapeutic effects of exercise (EX) versus resveratrol (RES) on steroidogenic and spermatogenic functions in streptozotocin (STZ)-induced T2DM in adult male albino rats receiving metformin (Met) treatment. Materials and Methods: The study was conducted on 48 rats equally divided into 6 groups (control, T2DM, Met, EX, RES and EX+RES). At the end of the study (8 weeks following T2DM induction), rats were sacrificed, semen was collected as well as blood sample for assessment of fasting glucose, insulin, testosterone, FSH and LH. Testicular tissues were collected for measurement of MDA, TAC and TNFα, gene expression of GDNF, histopathological examination and immunohistochemistry examination for Gfrα1 and c-RET. Results: STZ-induced T2DM revealed deterioration of rats’ spermatogenic function with decreased GDNF and Gfrα1 and c-RET expression in testicular tissue and also seminiferous tubules losing their normal structure. Increase in MDA and TNFα in testicular tissue with decrease in TAC. As well as disturbed HPG axis. Treatment with EX or RES alleviated the testicular functions, partially restored the histological features and increased GDNF, Gfrα1 and c-RET expression, notably, the combined therapy was better than the mono-therapy. Conclusion: EX and RES attenuate testicular dysfunctions in STZ-induced T2DM attributed to their antioxidant and anti-inflammatory effects together with overexpression of GDNF. Interestingly combined therapy showed a better improving effect compared to mono-therapy of either EX or RES.Summary: داء السكري هو متلازمة تمثيل غذائي، يتميز بارتفاع مزمن في سكر الدم ويمثل عبئا عالميا ضخما بسبب انتشاره المتزايد باستمرار، وله آثار تنتشر في جميع أجهزة الجسم ويعد تدهور الوظائف الإنجابية أحد أخطر مضاعفات داء السكري من النوع الثاني. ولهذا درسنا دور ممارسة التمارين الرياضة مقابل الريسفيراترول كونهما موادا مساعدة للميتفورمين في تحسين تدهور وظائف الخصية في الفئران المصابة بالنوع الثاني من داء السكري. كما قمنا بالتحقيق في الآثار المفيدة المحتملة للعلاج المشترك بممارسة التمارين الرياضية و الريسفيراترول في تنظيم حالة الالتهاب و الإجهاد التأكسدي الناجم عن النوع الثاني من داء السكري، بالإضافة إلى دورهما في التعبير عن عامل التغذية العصبية الدبقي GDNF المعروف بقدرته على تجديد الخلايا الجذعية المنوية. أجريت الدراسة على 48 من ذكور الفئران البيضاء البالغة بوزن يتراوح بين (180-200 جم).
