Introduction: The cultivation of hybridoma cells secreting monoclonal antibodies (MAbs) involves the use of media supplemented with fetal bovine serum (FBS). The serum provides essential nutrients for cell metabolism, growth, and differentiation. However its use is increasingly criticized for scientific, technical, and ethical reasons.This lead to widespread support of cultivation the hybridoma cells in serum free conditions. Methodology: In this work, we studied the effect of the serum supplemented, serum free and chemically defined media on the hybridoma cells growth characteristics and monoclonal antibody productivity. To protect the hybridoma cells, cryopreservation and revival under serum free conditions was assessed. Bambanker media was studied for cryopreservation of the cells at -80C instead of liquid nitrogen. Lab-scale production of MAbs and purification was done to diagnose circulation Schistosoma mansoni soluble egg antigen in patient serum by sandwich ELISA
